Analysis of Proteins of Mouse Sarcoma Pseudotype Viruses: Type-Specific Radioimmunoassays for Ecotropic Virus p30's

摘要: Murine sarcoma virus pseudotypes were prepared by infection of nonproducer cells (A1-2), which transformed the Gazdar strain mouse virus, with Gross (N-tropic), WN1802B (B-tropic), or Moloney (NB-tropic) viruses. The respective host range pseudotype viruses defined titration characteristics on appropriate Fv-1 genotype. Proteins from progeny analyzed sodium dodecyl sulfate-polyacrylamide gel electrophoresis. Bands present in both 65,000- and 10,000- to 20,000- molecular-weight regions distinguished their helpers. Furthermore, two protein bands noted p30 region murine (Gross), one corresponding p30, another slightly slower mobility. However, since mobility putative is nearly indentical that WN1802B, its presence could not be established Type-specific radioimmunoassays for applied analysis preparations, among several ecotropic tested, only homologous scored assay. By use these assays, found contain 8 48% helper-specific p30's; remainder presumably derived virus.
