A simple label-free electrochemical method for the detection of polynucleotide kinase activity by a peroxidase mimic: TiO2 nanotube array

摘要: Phosphorylation of DNA by polynucleotide kinase (PNK) plays an important role in a variety cellular processes, such as nucleic acid metabolism and damage repair. Strategies for simple effective PNK assay are urgent need. Here we report the development label-free electrochemical method based on mimic peroxidase character prepared TiO2 nanotube array (NTA) its specific attachment to phosphorylated DNA. The concentration T4 was successfully detected NTA electrode with detection limit 0.15 U mL−1. In process, only one-step incubation process used no labeling processes were involved. addition, treated foil anodized into it directly without any other complex modification which avoided leakage modifier, so excellent stability flexibility is worthy use real time monitoring.
