Identification of a unique epigenetic sub-microenvironment in prostate cancer

摘要: The glutathione S-transferase P1 (GSTP1) gene promoter is methylated in tumour cells more than 90% of prostate carcinomas. Recently, GSTP1 methylation was identified tumour-associated stromal addition to the epithelium. To define extent and location methylation, epigenetic mapping using pyrosequencing quantification an anatomical three-dimensional reconstruction entire human specimen with cancer were performed. Normal epithelium stroma, epithelium, laser capture-microdissected from multiple locations throughout gland. As expected, both normal distant not showed consistently high levels throughout. However, found be only a localized distinct sub-field tumour, revealing presence epigenetically unique microenvironment within cancer. Morphologically, consisted typical, non-reactive representing genomic alteration that appeared otherwise histologically normal. Similar control gland without low background all cell types specimen. These data suggest can occur sub-region may have implications for understanding biology clinical intervention.
