摘要: Matrix metalloproteinase activity was assessed in culture fluids of organ-cultured human skin by gelatin zymography. Both the 92-kD gelatinase/type IV collagenase and 72-kD were detected. Production enzyme substantially increased presence epidermal growth factor (EGF) hepatocyte (HGF) as compared to control but not insulin-like factor-1 (IGF-1) or keratinocyte (KGF). This is interest because our recent studies have shown that EGF HGF induce epithelial cells invade underlying stroma while normal architecture maintained IGF-1 KGF. Addition tissue inhibitor metalloproteinase-2 organ blocked expression active forms both enzymes concomitantly invasion. Epidermal keratinocytes, dermal fibroblasts endothelial grown monolayer examined for matrix production. The accounted most gelatinase fibroblast cell fluids. Increased production seen keratinocytes upon exposure factors induced invasion (EGF HGF) two did (IGF-1 KGF) much less effective. upregulated any four factors. Taken together, these data indicate epithelium under influence invasion-inducing contributes
