Analysis of P53 mutations and their expression in 56 colorectal cancer cell lines

摘要: A comprehensive analysis of the TP53 gene and its protein status was carried out on a panel 56 colorectal cancer cell lines. This based combination denaturing HPLC mutation screening all exons p53 gene, sequencing cDNA, assessing function by assaying induced expression phosphorylated p21 after exposing cells to γ-rays. In few cases where there no production message nor evidence functional protein, were sequenced directly. Thirteen lines had p53, 21 missense mutations (one which made detectable protein), 4 produced transcripts, remaining 18 truncating mutations. Thus, our results showed relatively high frequency (76.8%) in lines, with almost half being is rather higher such than usually reported.
