摘要: Abstract The conventional clotting methods for determination of plasma prothrombin all involve fibrinogen-fibrin conversion as an indicator thrombin activity. This is a complex reaction and preliminary attempts to follow the initial polymerization by rate analyzer (LKB 8600) were abandoned. It was more appropriate use synthetic chromogenic substrate (1). generated allowed split tripeptide yielding p-nitroaniline, liberation which followed in analyzer. In order convert we used reagent containing partially purified coagulation factors first phase. Other commercial reagents did not function this system. results are agreement with determinations normal individuals dicoumarol treated patients. Obvious advantages method attractive kinetic way assaying prothrombin. mechanized system gives better reproducibility saves manual work.
