摘要: The requirements for cholera toxin-catalyzed ADP ribosylation of the purified regulatory component adenylate cyclase are described. In addition to toxin, this reaction is dependent on or facilitated by NAD, GTP, phospholipid, and a factor found associated with plasma membranes from several sources. Factor activity heat-labile protease-sensitive but unaffected treatment N-ethylmaleimide. Gel filtration indicates that behaves as monodisperse species Stokes radius 3.2 nm. thus appears be protein distinct any known components cyclase. was also detected in cytoplasm S49 cells. cytoplasmic smaller (Stokes = 2.0 nm) than membrane-derived factor, it inactivated presence sodium cholate. initial rate activation toxin linearly related amount present reaction. This has allowed quantitation partial purification (33-fold detergent extracts) turkey erythrocyte membranes.
