POLYAMINES INCREASE IN SYMPATHETIC NEURONS AND NON- NEURONAL CELLS AFTER AXOTOMY AND ENHANCE NEURITE OUTGROWTH IN NERVE GROWTH FACTOR-PRIMED PC12 CELLS

摘要: Following axonal damage, sympathetic neurons are capable of regenerating and reinnervating their target tissues. Some years ago exogenous administration polyamines was shown to enhance this regeneration. Recently, it found that injury leads a dramatic up-regulation the expression arginase I in neurons. This enzyme catalyzes conversion arginine ornithine, which can subsequently be converted diamine putrescine and, ultimately, spermidine spermine. In present study, using an antiserum reacts with both spermine, we have increase polyamine levels non-neuronal cells superior cervical ganglion 2 5 days following transection ganglion's postganglionic trunks. Using PC12 primed nerve growth factor then stripped off culture dish replated as model system for axotomized neurons, treatment, or without factor, resulted increased percentage neurite whose length at least twice diameter neuron's cell body. These increases could seen within 48 h were still evident after 8 days. Together, these data support possibility endogenous involved normal regeneration occurs damage.
