The Pro12Ala and C–681G variants of the PPARG locus are associated with opposing growth phenotypes in young schoolchildren

摘要: Peroxisome proliferator-activated receptor γ is an important regulator of adiposity in mouse and man, common variation the PPARG gene has been associated with birthweight, adult obesity, insulin sensitivity type 2 diabetes. We hypothesised that these variants may be childhood obesity. Height weight were recorded for 2454 prepubertal children aged between 4 10 years, who then genotyped three locus: C–681G, Pro12Ala C1431T. No single variant was significantly height, or BMI. However, when modelling together we detected opposing interaction −681G Ala12 height weight, but not BMI (p=0.018, 0.013 0.119 respectively). The data consistent carriers being deficient energy storage/utilisation, leading to reduced growth. In contrast, variant, which increased accelerated two strong linkage disequilibrium. rare individuals bearing isolated demonstrated greatest from mean, most contrasting genotypes a 7 kg 6 cm standardised 7.4-year-olds (p=0.006 p=0.02 This study demonstrates quantitative trait analysis balance/growth locus complex requires use multiple genetic markers.
