Purification and characterization of an antitumor protein with deoxyribonuclease activity from edible mushroom Agrocybe aegerita

摘要: Scope Mushrooms are well known for their nutritional and medicinal value. Agrocybe aegerita has been used as a nutritious food around the world its herbal properties in Asia. In recent years, several antitumor proteins have identified from A. aegerita. The objective of this study was to purify novel protein aegerita. Methods results A deoxyribonuclease (AAD) purified through two-step chromatographic procedure shown possess activity against different cancer cell lines. Cells treated with AAD produced typical apoptotic morphological changes, which include chromatin condensation, accumulation sub-G1 cells caspase-8 cleavage. Biochemical characterization showed that it member DNase I family possessed divalent metal ion-dependent endonuclease activity. optimal temperature 50°C pH 8.5. MS-identified peptides were found match Unigene3821, 97% homology Aa-Pri1, our transcriptome. Conclusion We This fuller understanding would help us enhance use medical applications.
