摘要: Purpose Estrogen has been shown to have a modulating effect on cartilage thickness. This investigation was performed determine the effects of estrogen supplementation thickness, cellular proliferation, and type II X collagen production in skeletally mature rat cartilage, both an organ culture cell system. Materials Methods Mandibular condyles were harvested from 8-week-old female Sprague Dawley rats placed into tissue plates containing media with or without 17β-estradiol supplementation. Organ cultures labeled 5-bromo-2′-deoxyuridine day 2 4 mitotic index. Histomorphometric analysis sections used thickness (μm) various zones, as well total following exposure. Type immunohistochemically identified ECM hypertrophic chondrocytes using rabbit anti-rat antibody raised against NCl domain. The reaction visualized avidin-biotin peroxidase detection system (Vector Laboratories, Burlingame, CA). In separate experiment, articulating by collagenase digestion cultured at 5 × 105 cells per 35 mm plate. Second subculture divided groups: controls [10−8 M] (E2−10−8 M) grown confluence. establish growth curves for each group counts 2-day intervals. Results 17β-estradiol–treated had significant decrease after days (P Conclusion These data show that decreased inhibition chondrocyte proliferation increased maturation. observed showed potential role modulation cartilage.
