摘要: Publisher Summary This chapter discusses the advances in chromatography of lipids. The method preparation a lipid extract constitutes one most critical parts analysis by chromatographic methods. Extraction with organic solvents provides simple isolating all major phospholipids regardless their fatty-acid composition. isolation minor acidic phosphoinositol phosphatides (PtdInsPs) requires acidification, which causes destruction plasmalogens unless they are removed first. Good laboratory practice recommends that extractions be performed presence butylated hydroxy toluene (BHT) or other antioxidants, especially when working small amounts lipid. Because PtdInsPs and products hydrolysis degraded very rapidly, it is essential either to samples immediately after collection freeze situ store them at −70 o C. In fixation clamping microwave irradiation may helpful arresting degradation phosphate esters.
