Characterization and cell-free translation of mouse pituitary tumor messenger RNA which directs the synthesis of a corticotropin precursor

摘要: A wheat germ cell-free extract was used for the translation of poly(A)-containing RNA extracted from free and membrane-bound polysomes isolated AtT-20 mouse pituitary tumor. The mRNA associated with enriched by a factor 16 directing synthesis 31,000 molecular weight product which immunoprecipitated specific antibody against synthetic β 1–24 -corticotropin. Sucrose gradient analysis followed electrophoresis on formamide-polyacrylamide gels revealed that directs corticotropin-like has an approximate 400,000. Further evidence identity is provided presence common peptides in α-chymotryptic digests [ 3 H]lysine 35 S]methionine labeled synthesized products authentic corticotropin. Furthermore, under controlled tryptic digestion, both large corticotropin behave identical manner. These results indicate immunoreactive precursor to protein requires approximately 65% coding capacity molecule.
