Transfer of cyclosporine-associated syngeneic graft-versus-host disease by thymocytes. Resemblance to chronic graft-versus-host disease.

摘要: Syngeneic rat radiation chimeras treated transiently with cyclosporine (CsA) often develop a (GVHD-like syndrome after discontinuing the drug. CsA also casues medullary involution and loss of epithelium in thymus. Chronic graft-versus-host disease (GVHD), late occurring following bone marrow transplantation many features autoimmune diseases, is thought by ot result from thymic deficiency leading to failure specific tolerance for host. A direct connecton between chronic (GVHD) was tested transferring thymocytes CsA-treated syngeneic Lewis into irradiated secondary recipients. Nine 10 these recipients had evidence GVHD skin biopsies taken at 3 weeks posttranspant or autopsies 5 weeks. Changes included characteristic lichen planuslike infiltrates sclerodermalike changes skin, myositis tongue, hepatitis bile duct injury, interstitial ductal serous salivary glands. Immunoperoxidase stains tongue showed marked predominance W3/25+:OX8− lymphocytes. The hair follicles increased expression Ia antigen. thymus recipient variable thymocyte reconstitution cortex mild reduction relative size medulla. Stains cytokeratin moderate cortical total epithelium. These studies demonstrate that post-CsA resembling results an abnormal microenvironment. They provide additional linking GVHD.
