Spermine prevents cytochrome c release in glucocorticoid-induced apoptosis in mouse thymocytes.

摘要: Apoptosis can be induced in primary cultures of mouse thymocytes using the glucocorticoid dexamethasone. Addition polyamine spermine simultaneously with dexamethasone reduces induction apoptosis compared to treatment alone. We investigated signal transduction pathway at mitochondrial level order elucidate spermine's protective effect. Mitochondrial involvement is evident due loss transmembrane potential, release cytochrome c into cytosol and activation caspase-9 dexamethasone-treated thymocytes. The addition inhibited from mitochondria cytosol, also caspase-9. When mitogen concanavalin A (Con A) was added dexamethasone- plus spermine-treated thymocytes, number apoptotic cells pre-G1peak reduced treated only spermine. Comparing or showed a markedly latter.
