The dynein genes of Paramecium tetraurelia. Sequences adjacent to the catalytic P-loop identify cytoplasmic and axonemal heavy chain isoforms.

摘要: Paramecium tetraurelia is a unicellular organism that utilizes both axonemal and cytoplasmic dyneins. The highly conserved region containing the catalytic P-loop of dynein heavy chain was amplified by RNA-directed polymerase reaction. Eight different P-loop-containing cDNA fragments were cloned. Southern hybridization analysis indicated each fragment corresponds to separate gene there are at least 12 genes expressed in Paramecium. Seven eight cloned contain sequence motif A, which found dyneins, one contains B, dyneins from cell types do not have cilia or flagella. Two further investigated: DHC-6 DHC-8 B. Additional sequencing central portions these showed most closely matches sea urchin ciliary beta similar Dictyostelium. Deciliation cells resulted substantial increase steady state concentration mRNA but only small change mRNA. Antisera produced against synthetic peptides derived motifs A Competitive solid-phase binding assays demonstrated antiserum peptide-specific.
