Structural and functional characterization of paramecium dynein: initial studies.

摘要: Dynein arms and isolated dynein from Paramecium tetraurelia ciliary axonemes are comparable in structure, direction of force generation, microtubule translocation ability to other dyneins. In situ have dimensions substructure similar those Tetrahymena. Based on spoke arrangement intact axonemes, translocate axonemal microtubules sliding such that active (-) end directed motors the doublet which body cape binds (N) translocates adjacent (N + 1) tipward. After salt extraction, based ATPase activity, paramecium is found as a 22S 14S species. The three-headed molecule has unfolded dimensions; single headed. Both dyneins can be photocleaved by UV light (350 nm) presence Mg2+, ATP vanadate; photocleavage pattern differs seen with taxol-stabilized, bovine brain vitro. Under standard conditions, dynein, like organisms, at velocities about three times faster than dynein.
