Age-dependent accumulation of Dolichol in Rat Liver: is tissue dolichol a Biomarker of Aging?

摘要: Dolichols are long hydrophobic molecules broadly distributed in all tissues and cellular membranes of eukariotic cells. Dolichol affects membrane structure fluidity, membrane-associated protein activities, sensitivity to oxidative stress. Reports have shown that dolichols exhibit a remarkable (6- 30-fold) age-related increase the adult mature rats old flies, mice, humans. In our longitudinal study, accumulation dolichol was monitored liver tissue male Sprague Dawley fed ad libitum up age 27 months. addition 24-month-old subjected different regimens anti-aging diet restriction (40% calorie or every-other-day feeding libitum) were tested. A parallel study carbonyl (a proposed biomarker aging) made. addition, decline autophagy/proteolysis studied isolated cells, view essential role this function maintenance. Results show an age-dependent can be observed but not food-restricted rats, precedes altered proteins, is accompanied by autophagy. It concluded satisfies primary secondary applicable criteria desirable features required qualified as aging.
