摘要: The imbalance of Th1/Th2 subsets is an important pathogenic mechanism for insulin-dependent diabetes mellitus (IDDM). Calcitonin gene-related peptide (CGRP) has been found to play roles in the regulation T lymphocytes. We hypothesize that exogenous CGRP administration during insulitis may modulate balance Th lymphocytes, thereby providing a therapeutic intervention IDDM. established gene transfer by naked plasmid injection into skeletal muscles with electroporation enhancement. effect on pathogenesis IDDM was observedin autoimmune diabetic C57BL mice induced multiple low-dose streptozotocin (MLDS) administration. treatment significantly decreased morbidity diabetes, ameliorated hyperglycemia and insulin deficiency, inhibited lymphocyte infiltration islets, indicating protection beta cells against destruction. cell proliferation secretion Th1 cytokine IFN-gamma, increased level Th2 IL-10, but had no IL-4 TGF-beta1 secretion. also IL-12 IFN-gamma levels peritoneal effusion.
