摘要: This investigation represents the largest study so far published on human DNA damage and aging. The subject of this is damage, determined as alterations which give rise to complete molecular breaks in course treatment purified solutions with single-strand-specific nucleases. derived from milligram samples muscle individuals mostly undergoing surgical treatment. Care has been taken bring samples, once shut off blood circulation liquid nitrogen temperatures within few seconds. prepared by a procedure keeping handling DNAase attack low possible, however pushing purity, especially respect protein high possible. Highly treated way some sites are susceptible single-strand (ss) specific splitting (ss-events). Three different deoxyribonucleases have used: Nuclease S1, BAL31 Pea Endo-Nuclease. They very similar results, i.e. yield pieces given distribution. lengths these double-strand (ds) their electron microscopical pictures, either following image contours magnetostrictive stylo projected photo pad, mileage ruler, or integrating silver grains photo. weight averages ds threads between two ss-events for each individual 20 200 molecules. 470 contributing data were age groups 1 91 years. weights show considerable scatter an average 43.93 MDa standard deviation 17.99 MDa. Among (ssb) that split into such fraction, number increases highly significant fashion donor. From derives fact strand ssb decreases age. It remarkable significantly, too. On basis additional information mainly supplied donor himself his parents members main group M where grouped according life-style, into: (1) abstinent people, essentially non-smokers refraining use licit illicit drugs, sub-group N.(ABSTRACT TRUNCATED AT 400 WORDS)
