摘要: Abstract A sensitive ultrafast liquid chromatography coupled with triple quadrupole mass spectrometric (UFLC–MS/MS) method for the quantification of anhydrosafflor yellow B (AHSYB), a major active water-soluble pigment from Carthamus tinctorius , in rat plasma has been developed and validated. Sample preparation was achieved by protein precipitation four volumes methanol. Rutin used as internal standard (IS). The analytes were separated using C18 column an 8 min gradient elution, followed detection negative electrospray ionization (ESI − ) multiple reaction monitoring (MRM) mode. linear concentration range 25–10,000 ng/mL AHSYB. Intra-day inter-day precision variation less than 6.5%. relative error accuracy within ±9.4%. mean recovery AHSYB higher 70.9%. established successfully applied to pharmacokinetic study after intravenous (2.5 mg/kg) oral (30 mg/kg) dosing normal rats. And properties rats acute blood stasis differences between syndrome also investigated. results showed that compound poorly absorbed (∼0.3%) AUC 0– t ∞ F all significantly lower ( P
