摘要: A selective liquid chromatographic-mass spectrometric method has been developed and validated for simultaneous determination of senkyunolide I (SEI) H (SEH) from Chuanxiong Rhizoma in rat plasma. Plasma samples were extracted by liquid-liquid extraction with ethyl acetate separated on a Kromasil C18 column (250 × 4.6 mm, 5 µm), methanol-water (55:45, v/v) as mobile phase. The linear range was 0.05-25 µg/mL SEI 0.01-5.0 SEH, lower limits quantitation 0.05 0.01 µg/mL, respectively. Intra- inter-day precision within 10.0 9.8%, the accuracies (relative errors) <9.6 5.9%, mean recoveries 81.0-86.6 80.5-85.0% two anayltes, successfully applied to comparative pharmacokinetic study SEH normal migrainous rats after oral administration extract. results indicated that there obvious differences between behavior absorption significantly increased compared rats.
