Release of endotoxin from bacteria exposed to ciprofloxacin and its prevention with polymyxin B.

摘要: An in vitro system for studying the phenomenon of antibiotic-induced release endotoxin from gram-negative bacteria is described. Endotoxin was measured by a quantitative limulus lysate microassay. Using strain ofEscherichia coli,ciprofloxacin shown to cause an immediate and sustained increase compared control cultures whereas gentamicin did not, despite equally rapid bactericidal effect. The potential value anti-endotoxin effect polymyxin B examined using resistant ofKlebsiella aerogenes.Ciprofloxacin alone caused release, but when given with significantly less detected. These results indicate that rate cell death important than site antibiotic effect, suggest ciprofloxacin may act directly on wall as well DNA gyrase. It possible could be used reduce extent this would clinical benefit.
