Ultrasensitive detection of drug resistant cancer cells in biological matrixes using an amperometric nanobiosensor

摘要: Multidrug resistance (MDR) is a key issue in the failure of cancer chemotherapy and its detection will be helpful to develop suitable therapeutic strategies for patients overcome death rates. In this direction, we designed new amperometric sensor (a medical device prototype) detect drug resistant cells by sensing "Permeability glycoprotein (P-gp)". The probe fabricated immobilizing monoclonal P-gp antibody on gold nanoparticles (AuNPs) conducting polymer composite. relies sandwich-type approach using bioconjugate, where aminophenyl boronic acid (APBA) served as recognition molecule which binds with cell surface glycans hydrazine (Hyd) an electrocatalyst reduction H2O2 are attached multi-wall carbon nanotube (MWCNT) (APBA-MWCNT-Hyd). A linear range obtained between 50 100,000 cells/mL limit 23±2 cells/mL. proposed immunosensor successfully applied MDR (MDRCC) serum mixed samples. Interferences sensitive (SKBr-3 HeLa), noncancerous (HEK-293 OSE), other chemical molecules present real sample matrix examined. sensitivity excellent compared conventional reporter based assay.
