Genome-wide transcriptional analysis of T cell activation reveals differential gene expression associated with psoriasis.

摘要: Psoriasis is a chronic autoimmune disease in which T cells have predominant role initiating and perpetuating the inflammation skin. However, mechanisms that regulate cell activation psoriasis are still incompletely understood. The objective of present study was to characterize main genetic pathways associated with psoriasis. Gene expression profiles from vitro activated were obtained 17 patients 7 healthy controls using Illumina HT-12 v4 microarrays. From total 47,321 analyzed transcripts, 42 genes found be differentially expressed between (FDR p-value   1.2). Using an independent cohort 8 we validated overexpression SPATS2L (p-value =0.0009) KLF6 =0.0012) patients. weighted correlation analysis identified coexpression networks, also significantly (p-value < 0.05). Ontology allowed identification several biological processes each network. Finally, Set Enrichment Analysis over global transcriptome additional strongly (p-value < 0.0001). This has two new genes, KLF6, Functional analyses gene revealed results this provide important insight into biology common inflammatory disease.
