Inhibition of cryoprecipitation of murine IgG3 anti-dinitrophenyl (DNP) monoclonal antibodies by anionic DNP-amino acid conjugates

摘要: Previously we have demonstrated that eight out of nine IgG3 monoclonal antibodies (mAb) obtained from autoimmune MRL-lpr/lpr mice were able to self-associate and precipitate in the cold (Gyotoku et al., J. Immunol. 1987. 138:3785). To determine whether cryoprecipitation mAb is enhanced or inhibited presence specific ligand, established reactive with 2,4-dinitrophenol (DNP) hapten: four fusion spleen cells C57BL/6 immunized 2,4,6-trinitrophenylated keyhole limpet hemocyanin, three 129/Sv one BALB/c DNP-lipopolysaccharide. Five them induced cryoglobulins composed exclusively mAb. The binding negatively charged monomeric DNP-amino acid conjugates completely all five cryoprecipitating anti-DNP mAb, while incubation positively neutral had variable effects: increase, inhibition no change cryoprecipitation. In addition, induce non-cryoprecipitating Our data showed (a) derived non-autoimmune strains mice, similar an strain, possessed unique property form most cases; (b) although Fc-Fc interactions play a decisive role solubility, was markedly influenced after interacting their depending on charge hapten-amino conjugate. This suggested even minor interferences electrostatic equilibrium by hapten molecules dramatic changes solubility at low temperature.
