摘要: Bacterial and plasmid homo- heteroduplexes have been analyzed with a single-strand specific endonuclease, S1, of Aspergillus oryzae. Under appropriate assay conditions, there was high degree correlation between the deoxyribonucleic acid (DNA)-DNA homoduplex formation assessed by S1 endonuclease hydroxyapatite (HA). Heteroduplexes which contain extensive regions polynucleotide sequences in common are similarly recognized HA. In instances where is little or imperfect complementarity heterologous DNA strands, HA method give slightly different estimates. From duplex thermal stability experiments assayed preliminary evidence that well-matched identified enzyme not The permits an accurate, reproducible rapid determination sequence relationships may be seriously considered as choice for survey work investigations require large number DNA-DNA hybridization assays.
