Improved kinetic model for the transcutaneous measurement of glomerular filtration rate in experimental animals

摘要: Transcutaneous measurement of the glomerular filtration rate ( t GFR) is now frequently used in animal studies. GFR allows consecutive measurements on same animal, including multiple a daily basis, because no blood sampling required. Here we derive and validate a novel kinetic model for description transcutaneously measured FITC-Sinistrin excretion kinetics. In contrast to standard 1- 3-compartment models, our covers complete kinetic, injection distribution tracer plasma compartment. Because describes progression measurement, it further refinement by correcting baseline shifts observed occasionally during measurement. Possible reasons background signal include photo bleaching skin, autofluorescence, changes physiological state animals measurements, or effects arising from attachment device. Using new with modulated 3cp.b.m ), rats can reach comparable precision as those assessed using gold technique based constant infusion tracer. Moreover, variability simultaneous (parallel) well repeated animals, showed higher when was compared 1-compartment 1cp model.
