摘要: Abstract Uptake of 14 C-labelled amino acids into single oocytes was determined using 3 H-labelled choline to correct for extracellular space. Cycloleucine, a non-metabolisable acid sharing an entry mechanism with methionine and phenylalanine, transported in accord Michaelis-Menten kinetics. At levels below 8 mM, cycloleucine concentrated within the oocyte. The proportion sheep having functional transport system (i.e. flux > 1 nmole cm −2 h −1 ) highest pre-ovulatory follicles (97%), lowest atretic (59%). Amino fluxes germinal vesicle were similar at all stages development studied. An increase V max but not K m during meiotic maturation resulted doubling uptake metaphase II oocytes. These increased under gonadotropic regulation independent nuclear maturation. by mouse approximately half that measured
