摘要: A number of studies have demonstrated that malignant transformation is associated with an increase in glycolytic flux and anaerobic aerobic cellular lactate excretion. Using quantitative bioluminescence imaging various primary carcinomas patients (uterine cervix, head neck, colorectal region) at first diagnosis the disease, we showed concentrations tumors vivo could be relatively low or extremely high (up to 40 micromol/g) different individual within same lesion. In all tumor entities investigated, molar were correlated a incidence distant metastasis already early stage disease. Low ( approx. 8 micromol/g). Lactate dehydrogenase was found upregulated most these compared surrounding normal tissue. Numerous recent reports support data by demonstrating biological activities can enhance behavior cancer cells. These mechanisms include activation hyaluronan synthesis tumor-associated fibroblasts, upregulation VEGF HIF-1 alpha, direct enhancement motility which generates favorable conditions for metastatic spread. Thus, accumulation not only mirrors but also actively enhances degree malignancy. We propose determination may serve as basis novel metabolic classification lead improvement prognosis therapy clinical oncology.
