Analysis of Nicotine, 3-Hydroxycotinine, Cotinine, and Caffeine in Urine of Passive Smokers by HPLC-Tandem Mass Spectrometry

摘要: Background: A method is described for the simultaneous analysis of nicotine and two its major metabolites, cotinine 3-hydroxycotinine, as well caffeine from urine samples. The was developed to assess exposure restaurant hotel workers environmental tobacco smoke. Methods: includes sample pretreatment reversed-phase HPLC separation with tandem mass spectrometric identification quantification using electrospray ionization on a quadrupole ion trap analyzer. Sample followed standard protocols, including addition base before liquid-liquid partitioning against dichloromethane solid matrix, evaporation organic solvent gaseous nitrogen, transferring vials buffer. run on-line ionization-tandem detection. Results: detection limits procedure were in 1 μg/L range, except (10 urine). Still lower can be achieved larger volumes. Recoveries treatment varied 99% (cotinine) 78% (3-hydroxycotinine). Conclusions: straightforward not labor-intensive and, therefore, permits high throughput samples excellent prospects automation. applicability demonstrated small-scale study employees.
