摘要: The potential to block tumor growth by inhibition of the neoangiogenic process represents an intriguing approach treatment solid tumors. high proliferation rate in deprived proper vascularization would be balanced cell death due lack diffusion nutrients and oxygen. Matrix metalloproteinases (MMPs), angiogenic factors, their receptors are main targets increasing number clinical trials approved test tolerance therapeutic efficacy antiangiogenic agents. We observed that a series substances proposed as possible cancer chemopreventive agents show properties when tested vitro vivo angiogenesis models. demonstrated N-acetyl-l-cysteine is able reduce invasive metastatic melanoma cells, inhibit endothelial invasion direct MMP activity. also showed epigallocatechin gallate (EGCG), flavonoid from green tea possesses activity experimental epidemiological studies, potent inhibitor MMP-2 MMP-9. Angiogenesis has been target for nonsteroidal anti-inflammatory drug Based on these data, we hypothesize other agents, including natural or synthetic retinoids, steroid hormone antagonists, peroxisome proliferator-activated receptor gamma ligands, vitamin D, protease inhibitors, might have antiangiogenesis important mechanism action, novel concept will term 'angioprevention'. analyze mechanisms how why could exert effects aimed at controlling growth, use clinic.
