Prolonged treatment schedules with intraperitoneal 5-fluorouracil diminish the local-regional nature of drug distribution.

摘要: The levels of 5-fluorouracil (5-FU) in plasma and peritoneal fluid were determined after the bolus administration drug 2 L dialysis through a Tenckhoff catheter into cavity colon cancer patients. An increasing dose intraperitoneal (i.p.) 5-FU resulted an increased level blood. In 18 treatment cycles studied four patients, clearance from by day 5 i.p. treatment. one these did not further increase between days 8 12. If ascites was removed prior to subsequent administration, absorption markedly decreased. These studies show that given within body compartment may profoundly affect pharmacokinetics doses same chemotherapy.
