摘要: The novel sodium channel PN3/alpha-SNS, which was cloned from a rat dorsal root ganglion (DRG) cDNA library, is expressed predominantly in small sensory neurons and may contribute to the tetrodotoxin-resistant (TTXR) current that believed be associated with central sensitization chronic neuropathic pain states. To assess further role of PN3, we have used electrophysiological, situ hybridization immunohistochemical methods monitor changes TTXR distribution PN3 normal peripheral nerve-injured rats. (1) Whole-cell patch-clamp recordings showed there were no significant TTX-sensitive densities DRG after constriction injury (CCI) sciatic nerve. (2) Additionally, change expression mRNA up 14 d CCI. not detected sections brain spinal cord taken either or (3) In contrast, studies major subcellular protein caused by CCI complete transection intensity immunolabeling decreased increased nerve axons at site injury. alteration attributed translocation presynthesized, intracellularly located neuronal somata axons, subsequent accumulation specific redistribution an important factor establishing hyperexcitability resultant pain.
