Expression of Granulocyte-Macrophage Colony-Stimulating Factor Receptors in Human Prostate Cancer

摘要: We studied the expression and function of granulocyte-macrophage colony-stimulating factor (GM-CSF) receptor in human prostate carcinoma cell line LNCaP looked for its presence normal neoplastic prostatic tissue. The GM-CSF is composed two subunits, alpha beta. While isolated subunit binds at low-affinity, beta does not bind by itself; but complexes with to form a high-affinity receptor. Quantitative reverse transcriptase-polymerase chain reaction (RT-PCR) showed mRNAs encoding subunits cells, proteins was confirmed immunolocalization anti-alpha anti-beta antibodies. Receptor binding studies using radiolabeled that cells have about 150 sites kd 40 pmol/L approximately 750 low-affinity 2 nmol/L. signaled, time- dose-dependent manner, protein tyrosine phosphorylation induced proliferation cells. Immunolocalization low level tissue, substantial benign hyperplasia prominent Maximal both observed carcinomas metastatic lymph node bone. Tumor stained positively antibodies were also reactive antibodies, indicating they express receptors. Our data show functional receptors expression. These findings imply hyperplastic tissues may be responsive GM-CSF.
