摘要: To investigate the effect of H-2Kd gene on lymphocyte membrane, we constructed a small interfering RNA (siRNA) that targets and compared cytotoxicity mouse lymphokine-activated killer (LAK) cells with different expression states. H-2Kd-targeting siRNA was transfected into spleen lymphocytes BALB/C mice. Flow cytometry (FCM) then performed to examine in control cells. Additionally, toward H22 K562 cell lines evaluated vitro using LDH release assay. significantly reduced levels target protein, whereas pure transMessenger non-silencing did not inhibit at concentrations tested. The siRNA-treated LAK significantly. knockdown by may be associated neoplasm lines.
