Crocin antagonizes ethanol inhibition of NMDA receptor-mediated responses in rat hippocampal neurons

摘要: We have previously found that crocin (crocetin di-gentiobiose ester) antagonizes the inhibitory effect of ethanol on long-term potentiation in rat hippocampus vivo and vitro. To explore mechanisms underlying antagonism against ethanol, we investigated effects synaptic potentials mediated by N-methyl-d-aspartate (NMDA) receptors dentate gyrus hippocampal slices. Synaptic potential non-NMDA was recorded normal medium (1.3 mM Mg2+), while NMDA receptor-mediated isolated low (0.13 mM) Mg2+ containing receptor antagonist 6-cyano-7-nitroquinoxaline-2,3-dione (10 microM). Crocin microM) alone did not affect nor receptors. Non-NMDA response slightly inhibited 100 selectively lower concentrations (10-50 ethanol. inhibition but significantly blocked 10-50 In addition, performed whole-cell patch recording with primary cultured neurons, confirmed inward currents evoked application NMDA. These results suggest specifically responses neurons.
