New Methods for Detecting DNA Sequence Variation in Relation to Aging

摘要: The availability of accurate and sensitive methods for the detection DNA sequence variation is an important prerequisite obtaining insight in genetics aging. changes somatic cells are considered as causal factors aging, while accumulation germ line late-acting deleterious mutations could underly genetic component age-related disorders, such Alzheimer disease. We have taken two new experimental approaches studying genomic from higher animals. First, we used transgenic mice harboring a number tandemly integrated shuttle vectors with lacZ bacterial marker gene measuring both spontaneous induced various organs tissues. Second, by using two-dimensional electrophoretic separation system (size-separation neutral gels, followed denaturing gradient polyacrylamide gel on basis content) been able to detect minute variations minisatellite simple sequences. use these novel systems molecular aging will be discussed.
