Cloning, expression and functional characterization of the polyunsaturated fatty acid elongase ( ELOVL5 ) gene from sea cucumber ( Apostichopus japonicus )

摘要: Long chain polyunsaturated fatty acid (PUFA) are beneficial for maintaining the health, growth and development of an organism could reduce risk some diseases. The ability to endogenously produce PUFA, especially in invertebrates, is largely unknown. To study function elongase genes PUFA biosynthesis Apostichopus japonicus, we cloned ELOVL5 homology gene from intestinal cDNA A. japonicus (Aj-ELOVL5). Aj-ELOVL5 encoded a 318 amino (AA) protein that exhibited all characteristics family, such as histidine box motif four putative transmembrane-spanning domains. results tissue expression profile revealed body wall highest level compared with other adult tissues. We also found enzyme elongate γ-linolenic (18:3 n-6) eicosapentaenoic (20:5 n-3) dihomo-γ-linolenic (20:3 docosapentaenoic (22:5 n-3), respectively. Our indicated had capacity biosynthesize C18/C20 substrates.
