Combining capillary electrophoresis matrix-assisted laser desorption/ionization mass spectrometry and stable isotopic labeling techniques for comparative crustacean peptidomics

摘要: Herein we describe a sensitive and straightforward off-line capillary electrophoresis (CE) matrix-assisted laser desorption/ionization mass spectrometry (MALDI-MS) interface in conjunction with stable isotopic labeling (SIL) technique for comparative neuropeptidomic analysis crustacean model organisms. Two SIL schemes, including binary H/D formaldehyde novel, laboratory-developed multiplexed dimethylated leucine-based isobaric tagging reagents, have been evaluated these proof-of-concept experiments. We employ techniques CE-MALDI-MS quantitative peptidomic analyses of the pericardial organs isolated from two species, European green crab Carcinus maenas blue Callinectes sapidus. Isotopically labeled peptide pairs are found to co-migrate CE fractions changes relative abundances obtained by comparing peak intensities respective pairs. Several neuropeptide families exhibit response salinity stress, suggesting potential physiological functions signaling peptides.
