摘要: Elafin is an elastase-specific inhibitor with increased transcription in normal mammary epithelial cells compared to carcinoma cells. In this report, we test the hypothesis that inhibition of elastase, through induction elafin, leads human breast cancer cell viability and, therefore, predicts survival patients. Panels and immortalized cells, along were used examine impact adenoviral-mediated elafin expression or shRNA-mediated elastase on growth xenografts nude mice. To determine prognostic significance decreased patients invasive cancer, previously published gene array datasets interrogated. had no effect non-tumorigenic but resulted marked lines. Control-treated generated a tumor burden necessitated sacrifice within one month initial treatment, whereas xenograft-bearing mice treated Ad-Elafin alive at eight months reduction growth. Elastase mimicked these results, showing vitro vivo. Low correlated significantly reduced time relapse, when combined high was associated Our data suggest plays direct role suppression tumors thus serves as indicator for
