摘要: Cell-matrix interactions and the ordered deposition of basement membrane (BM) components are major importance for maintenance tissue homeostasis in complex epithelia. This aspect was studied vitro a coculture system designed as an oral mucosa model. As crucial epithelial features kinetics proliferation, expression site-specific keratins well integrin patterns correlation to synthesis BM were assessed by immunohistochemistry situ hybridization. Comparison with non-cornified gingiva origin revealed different stages development, eventually leading complete reconstruction within time frame 1–3 weeks. Second, after 2 weeks improvement histoarchitecture correlated predominant extension K13 suprabasal cell compartment, integrins α3β1 α6β4 including their ligand laminin-5 (d) accumulating components. Third, virtually normalization at 3 indicated restriction area, regular localization K13, polarization parabasal cells linear codistribution collagen IV, “classical” laminin (-1 or -10) underneath cells. Thus, these organotypic cocultures represent relevant equivalents non-keratinized typical gingival differentiation addition suitable models preclinical trials such prospective dental material testing.
