摘要: The wild-type E6 and E7 genes of human papillomavirus type 16 (HPV16) can cooperate to immortalize normal keratinocytes in culture. open reading frame HPV16 other HPV types highly associated with cervical cancer has the potential encoding both full-length two truncated E6* proteins, latter being generated via splicing within portion E6-E7 polycistronic transcript. Those types, such as HPV6, that are infrequently carcinoma lack splice site encode only a E6. We have now found that, addition cooperating keratinocytes, induce anchorage-independent growth NIH 3T3 cells trans-activate adenovirus E2 promoter. HPV6 was also able promoter, although it inactive cell transformation assays. An mutant which expressed active all three assays, indicating proteins not required for these activities. plasmid encodes did potentiate activity mutant. mutation prevented mRNA severely reduced level protein increased protein. Taken together, results suggest primary function is facilitate translation reduce E6, rather than generate biologically proteins.
