摘要: Transient transfection into L8 myoblasts has been used to study the rat alpha-actin gene promoter. Demodification of specific sites occurs in two stages, with a hemimethylated intermediate formed within few hours after entry construct cell. The removal methyl moiety from complementary strand takes place delay at least 48 hr, and both events are actively carried out absence DNA replication. By assaying activity during course transfection, it was possible demonstrate that demethylation strands critical CpG loci is essential activate transcription. Genetic analysis revealed existence cis-acting elements required for demethylation. recognition these early differentiation process probably leads demodification make accessible its transcription factors.
