摘要: The cytosolic phosphenolpyruvate carboxykinase [PEPCK; GTP:oxaloacetate carboxy-lyase (transphosphorylating), EC 4.1.1.32] gene was isolated from a rat genomic library, and map of the methylatable sites C-C-G-G G-C-G-C has been constructed. extent methylation 18 in PEPCK adult liver, kidney, spleen, heart muscle fetal liver analyzed using 5-methylcytosine sensitive enzymes Hpa II Hha I. This analysis revealed extensive undermethylation kidney (PEPCK-expressing tissue), whereas spleen as well (PEPCK-nonexpressing tissues) heavily methylated. However, unlike nonexpressing tissues, region middle found to be partially hypomethylated liver. hypomethylation correlates with competence expressed. Treatment fetuses by utero injection 5-azacytidine causes hypomethylation-associated activation gene. Taken together, present findings suggest sequential loss methyl groups during development. When related expression, demonstrates an early stage prior transcription characterized discrete later developmental all associated mature active around time birth.
