摘要: ♦ Bac kground: Dial ysis patients are classified according to their peritoneal permeability as low transporter (LT, solute permeability) or high (HT, permeability). Tight junction (TJ) proteins critical maintain ions, molecules and water paracellular transport through peritoneum. Exposure dialysis solu tions causes damage TJ in human mesothelial cells (HPMCs). We analyzed the quantity, distribution function of proteins: claudin-1, -2 -8, ZO-1 occludin, HPMC cultures from LT HT patients. Since all-trans retinoic acid (ATRA) might modify expression proteins, we studied its effect on HPMCs. Me thods: Control HPMCs were isolated omentum, while obtained dialy sis effluents. Cells cultured presence ATRA 0, 50 100 nM. Transepithelial electrical resistance (TER) measurement, immunostaining Western blot analyses performed. R esults: exhibited lower TER than control monolayers. Immunofluorescence for was weak discontinuous along cell contour, HT. Furthermore, claudin- 1, occludin expressions decreased. In all groups, claudin-2 localized at nuclei. observed that improved increased This retinoid did not -8 expressions. All-trans decreased HT, but had no LT. Conclusions: T ight junctions altered dialyzed The monolayer has LT, which be associated with these a therapeutic alternative integrity, since it localization expression.
