摘要: Measuring intact human parathyroid hormone (hPTH) still presents many problems. Owing to the rapid uptake and cleavage of hPTH(1–84) by liver, PTH is removed from circulation within minutes [17]. As a consequence, plasma concentrations are very low. RIAs directed against primary region (residues 28–48) appeared be specific for but were usually not sensitive enough detect in all healthy persons [15]. To obtain necessary assay sensitivity, Lindall et al. [14] used an extraction step: columns with solid-phase-bound anti-N-terminal antibodies extract concentrate N-terminal fragments samples. The was finally detected subsequent RIA midregion fragments.
