摘要: We have previously established that eosinophils studied ex vivo from the sputum of asthmatics express intercellular adhesion molecule-1 (ICAM-1) and HLA-DR, whereas peripheral blood do not these surface proteins. On incubation highly purified (greater than 99.5% pure) normal subjects with T cell supernatants, eosinophil ICAM-1 was induced in 24 h, HLA-DR maximally within 48 h. Recombinant cytokines enable survival (IL-5, IL-3, granulocyte macrophage-CSF) were found to be unable induce or even when pooled at concentrations individually required for survival. However, synergy between factors TNF (-alpha -beta) mainly responsible induction, IL-3 IFN-gamma occurred induction. Culture presence cycloheximide prevented expression showing de novo protein synthesis is occurring. At a functional level we demonstrate ICAM-1-bearing increased capacity autologous cells. In contrast, HLA-DR-expressing mediated Ag-specific proliferation an HLA-DR-restricted clone inhibitable by anti-HLA-DR anti-ICAM-1 mAb. Since eosinophil-mediated Ag presentation treatment glutaraldehyde chloroquine, this suggests participate uptake, processing, accessory functions. Thus, through induction on tissue eosinophils, interact leukocytes present
