摘要: Polyamine contents increase during epithelial carcinogenesis in both rodents and humans. The activity of ornithine decarboxylase (ODC), the first enzyme polyamine synthesis mammals, is increased cancers, ODC has become a target for cancer chemoprevention. Difluoromethylornithine (DFMO) specific inhibitor under evaluation clinical trials prevention including those colon, prostate, skin. Tissue, serum, other bodily fluid endpoints have been used to monitor effects DFMO human chemoprevention trials. Cell tissue are regulated by mechanisms influencing synthesis, catabolism, uptake, efflux. These regulatory include transcriptional posttranscriptional processes. Tissue measurements often failed be reliable measures either or response DFMO. Levels polyamines less subject measurement variability than activity, closely associated with administered dose randomized Target validated biomarkers biochemical efficacy There need new predictive agents that affect metabolism could measured sources tissues.
