NMR triple-quantum filtered relaxation analysis of 17O-water in insulin solutions: an insight into the aggregation of insulin and the properties of its bound water.

摘要: Abstract Transverse triple-quantum filtered NMR spectroscopy (TTQF) of 17 O-water was used to study the properties water in insulin solutions at different Zn 2+ concentrations and pH values. It established that strongly bound molecules are already present Zn-free insulin. On assumption effective correlation time a molecule, τ sb , is 10 ns, apparent number ∼3 4 per monomer. Addition equivalent ∼2 g-atoms hexamer did not produce substantial increases overall TTQF signal intensity fraction water. The dramatic enhancement signals observed for samples with /hexamer ratio greater than ∼2:1 could be attributed increase water, due formation higher-order oligomers protein.
